DNA Decoded: Why Test Your Oldest Relatives First?

When family historians decide to take a DNA test, the first instinct is often to test themselves. That can be useful—but if an older parent, aunt, uncle, grandparent, or great-aunt is willing and able to test, that person is usually the higher-priority tester.

The reason is inheritance. Autosomal DNA is reshuffled at every generation. You receive roughly half of your autosomal DNA from each parent, but not a perfectly representative half of every earlier ancestor. Some segments your parent inherited from a grandparent were not passed to you at all. An older relative therefore carries ancestral DNA that younger descendants may never have received.

Imagine that your grandmother and her sister inherited different combinations of DNA from the same parents. Your parent inherited only part of your grandmother’s combination, and you inherited only part of your parent’s. Testing your grandmother—or her sibling—lets you reach closer to the generation you are researching and may produce matches or segments that are invisible in your own results.

Older relatives also make match interpretation easier. If your mother tests, matches shared with her can be assigned to your maternal side. If her oldest brother also tests, comparing their match lists may reveal relatives connected through different portions of their parents’ DNA. Testing siblings is not redundant; each sibling is a different genetic sample of the same parents.

The same principle can help with brick walls. If you are trying to identify the parents of a great-grandmother, test the living relative who is most closely related to her through the fewest generations. A grandchild of that ancestor generally has more relevant DNA than a great-great-grandchild. Closer testers tend to share larger, easier-to-analyze amounts of DNA with useful cousins.

Start with a testing plan rather than sending kits at random. Write down the research question, identify every living descendant of the target ancestral couple, and rank candidates by generational closeness. Consider testing relatives from different children of the target couple, because their results can help separate matches associated with one branch from matches associated with another.

Consent must come first. Explain what the test may reveal, including unexpected parentage, unknown relatives, or family secrets. Discuss privacy settings, data sharing, law-enforcement matching policies where applicable, who will manage the account, and whether the tester wants continuing access. Never pressure a relative. A clear “no” should remain a no.

If a relative agrees, document the process. Record the testing company, kit number, account owner, manager, date collected, and the tester’s relationship to the family. Build or link a small, accurate family tree so matches have context. Save permitted result files and reports in a secure, organized location, following both the company’s terms and the tester’s wishes.

Different DNA types answer different questions. Autosomal DNA is the broadest choice for recent cousin matching across family lines. Y-DNA follows a direct paternal line and requires an appropriate male-line tester. Mitochondrial DNA follows a direct maternal line and can be tested by anyone who inherited it, although only daughters pass it forward. The oldest-relative principle still applies, but the correct tester depends on the lineage and question.

DNA testing cannot replace documentary research. Names, places, timelines, records, and carefully built trees are what turn a match into evidence. Yet the opportunity to test an older generation may be temporary. When a willing older relative is available, preserving that genetic evidence can be one of the most valuable steps a family historian takes.

Oldest-Relative Testing Priority

  1. Define the ancestor or relationship you want to investigate.

  2. List living relatives closest to that ancestor.

  3. Prioritize the oldest generation and useful siblings or cousins from separate branches.

  4. Discuss informed consent, privacy, surprises, access, and account management.

  5. Choose the DNA test that fits the research question.

  6. Connect results to a sourced family tree and document the testing details.
